When I went through the local cartel wanted $50/ head for line handing and you need four of them.
I was able to swap with a NZ boat two from my boat and two from theirs. We took their boat through in one day starting at 6 AM at the first dock. The pilot came aboard at 5 AM. We got a bus back to Colon that night after dinner in Panama City ( they go every hour and cost about $1 each)
The NZ crew arrived back in a few days for my transit.
You need four 100'+ long 3/4" line to keep the boat in place. They must reach up to the lock sides. They can be hired but I used two anchor lines from my boat and two from the NZ boat for both transits. You also need a lot of fenders. The Panama canal YC in Colon had some covered car tires for free loan.

I stayed in Colon at the PCYC marina at $16 /day but my friends moored out on the "flats" for free and dingied in each day. I waited 5 days to get through.
You have to do a lot of paper work. Immigration, customs, cruising permit, Canal transit permit and arrange for pilot and transit booking. Some people pay agents $250 to help organise all this.
It is going to take time any way so I think spending a day your self is worth it. I used a Taxi driver named Carlos Solano (Taxi 3T-518 phone 445 1240 or 445 1608) He was very good and spent the day with me as driver, interpreter and negotiator for $50 We got it all done in 6 hrs on a Saturday.
If I could not find a yachty to she line handling with I would try organising some local me from out side the YC through the taxi driver who would be happy to work for $10/day.
In Panama city I stayed at The Belboa YC (Club house now burnt down). Same price a PCYC but no marina. That is worth it for a few days to clear out and have a look around. If you want to stay longer you can find places to anchor free. I anchored the other side of the canal peninsula and left by dingy with a cafe that had baddle boats. There is a lot to see in Panama. May boaties just use the canal and sail of and miss this largely undeveloped tourist free beautiful country.

I'm hot on your heels David! I'm planning on canal transit and
leaving panama for Galapagos about the same times as you.

Keep an eye out for me. I'll be on a light blue Allied Princess
Ketch. The Lucky Dolphin. Maybe we can help each other out getting
through the canal. I know I'll need line handlers and was planning
on loaning myself out to line handle on someone else's boat before I
  take mine through anyway.

I'll be leaving Florida on or about the 1st of November.

  Happy cruising.
